i use both in the same sentance frequently. If something is said to me that i find wicked surprising, i usually say "Nuh-uh...NO SUH!?" Basically meaning "What!? No Way!" The three ...'s means there is a pause in between to two...
yes-suh, no-suh. taken from the way southern blacks speak (still to this day): yessuh (massuh). nossuh (massah). some times i think it's disgusting so i'll just use 'NOOO!' to mean the same thing.yuh-huh and nuh-uh are just too fun not to use in the most exaggerate way possible.
I disagree that "no suh" has any connection to southern blacks saying "yessuh" or "nossuh." "No Suh" is just a very idiosyncratic expression that means "That's amazing" or "I don't believe that."
Said this once shortly after moving from Boston to Indiana. Never occurred to me that it wasn't regional. Had some very confused Hoosiers on my hands and haven't used the phrase since.

re: No SUH!
is it the same as nuh uh?
re: No SUH!
No, "nuh-uh" is not the same as "no suh"."nuh-uh" simply means "no"; "no suh" means "no sir", in effect, "no way" and can also be used sarcastically
re: No SUH!
It is for "no, sir" with the accent applied "no, suh"
